Gene delivery to neurons of the dorsal root ganglia using adeno- associated viral vectors

چکیده
Viral vector-mediated gene transfer, especially using adeno-associated viral (AAV) vectors, is a powerful strategy to manipulate gene expression in vivo in injured neurons. In this chapter we provide two methods to efficiently transduce dorsal root ganglion (DRG) neurons in vivo using AAV. We describe detailed procedures to perform direct injections into specific DRG and delivery via the intrathecal space to transduce the lumbar DRG. Finally, we discuss the specific advantages and disadvantages of these two methods of delivery. The main advantages of direct injection are that high transduction rates can be achieved in specific ganglia (L4/L5) with low amounts (µl) of a viral vector stock, however the procedure is complex, invasive and time consuming. Intrathecal injection has the advantage of being a fast and simple method to transduce multiple DRG bilaterally, and involves no surgical manipulation of the DRG. However, intrathecal delivery does require much larger amounts of viral stock (10 to 20µl), and has the disadvantage that viral particles will leak from the cerebrospinal fluid to the spinal cord and/or peripheral tissues.

P164: Adeno-Associated Viral Vectors in Duchenne Muscular Dystrophy
Duchenne muscular dystrophy (BMD) is an inherited X-link disease. The incidence of this muscle-wasting disease is 1:5000 male live births. Mutation in the gene coding for dystrophin is the main cause of BMD. Most cases of this disease succumb to respiratory and cardiac failure in 3rd to 4th decades. The slow progression of BMD and recent achievement of gene therapies make it as an appropriate c...

BACKGROUND Neuronal transduction by adeno-associated viral (AAV) vectors has been demonstrated in cortex, brainstem, cerebellum, and sensory ganglia. Intrathecal delivery of AAV serotypes that transduce neurons in dorsal root ganglia (DRG) and spinal cord offers substantial opportunities to 1) further study mechanisms underlying chronic pain, and 2) develop novel gene-based therapies for the tr...

There is increasing trend in using recombinant stem cells as novel therapeutic candidates in different diseases. These studies encompass different applications from targeted homing of Mesenchymal Stromal (stem) Cells (MSC), to arming them with different cytokines. متن کاملIn Vivo Gene Knockdown in Rat Dorsal Root Ganglia Mediated by Self-Complementary Adeno-Associated Virus Serotype 5 Following Intrathecal Delivery
We report here in adult rat viral vector mediate-gene knockdown in the primary sensory neurons and the associated cellular and behavior consequences. Self-complementary adeno-associated virus serotype 5 (AAV5) was constructed to express green fluorescent protein (GFP) and a small interfering RNA (siRNA) targeting mammalian target of rapamycin (mTOR). The AAV vectors were injected via an intrath...

The key principle of gene delivery to articulations by direct intra-articular injection is to release complementary DNA(cDNA)-encoding medical products that will lead to maintained, endogenous production of the gene products withinthe articulation. In fact, this has been accomplished for both in vivo and ex vivo gene delivery, using several vectors, genes, and cells in some animal models. Some c...
